昭和初期の大阪。北浜の株仲買商の娘として生まれた浦野みお(藤吉久美子)は、走る能力を見出されオリンピックへの夢を抱く。しかし、実家が倒産したことで夢を断念。道頓堀の芝居茶屋の女将・時田せい(南田洋子)に頼み、住み込みのお茶子として働き始める。その後、舞鶴の駅弁店で働き、結婚、出産。そして戦後再び道頓堀で店を開き、人生のマラソンを走り抜く。なにわ女のみおつくし半生記。
